Research Description

Dr. Cote's major areas of investigation are detection and characterization of micrometastases in patients with epithelial tumors and analysis of cellular and molecular markers of tumor progression in patients with cancer. He is focused on the study of bladder, breast, lung and prostate cancer. His work includes an examination and dissection of the relationship of cell cycle abnormalities and cancer progression, and an investigation of the clinical significance of angiogenesis in cancer prognosis.

Other major projects that he is involved in are: Analysis of tumor suppressor genes in human cancer; Molecular Mechanisms of Racial/Ethnic Variation in Prostate Cancer Risk and Mechanisms of Androgen Independence in Prostate Cancer

In addition, Dr. Cote is interested in the development and application of methods to detect occult micrometastasis in the bone marrow and lymph nodes of patients with early stage and late stage breast, lung, prostate and colon cancer using immunohistochemistry and the reverse transcription polymerase chain reaction techniques. The presence of occult metastasis may be an important means to identify patients at greatest risk for developing metastatic disease. He is also investigating methods by which the detection of occult micrometastasis can be automated using image analysis.

Dr. Cote has been involved in various collaborative works, one with Drs. Taylor and Shi, on the investigation of techniques in immunohistochemistry, particularly the antigen retrieval technology. The other is in collaboration with other researchers from USC, UC Berkeley and Oak Ridge National Laboratory, which we are developing an optomechanical chip for specific detection of molecular interactions relevant to cancer. They are expected to provide detection and quantitative analysis of molecular interaction between DNA, RNA and protein molecules in vitro. These portable microarrays will be used in cancer diagnosis and prognosis.
