Activin-mediated inhibition of prostate cancer cell growth and its gene regulation
Activin is a growth factor that modulate the growth of several types of tumor cells, including human prostate cancer cells. We have previously shown that activin is produced and the mRNAs encoding activin and its receptors are expressed in the intact rat prostate gland as well as a human prostate cancer cell line, LNCaP. Exogenously administered recombinant human activin inhibits cell proliferation and enhances apoptosis in LNCaP cells. We hypothesized that activin/activin receptor complex may mediate expression of a series of apoptosis-related oncogenes resulting in the inhibition of cell proliferation in LNCaP cells.
To elucidate the mechanism whereby activin regulates the uncontrolled proliferation of cells in prostate cancer, we determined whether activin inhibits cell proliferation and promotes apoptosis as well as oncogenes in cell growth and prostate markers in LNCaP cells. We have developed two simple, rapid subtractive hybridization methods (USC&HSC) and isolated several up- and down- regulated genes.
Finally, we are exploring the hypothesis that oncogenes, prostate markers, growth promoting factors and androgens are involved in the activin-mediated inhibition of prostate cell growth as well as in human advanced prostate cancer. To this end, we developed techniques to generate full-length DNA libraries from less than 20 cells of tissue sections and identified the up- and down- regulated genes in these tissues.
