The Center's Core facilities provide key services to support integrated Center investigations.

ANIMAL CORE SERVICES

This core provides the rat intragastric ethanol infusion model as well as animal models for cirrhosis which will be used by all Center investigators. It also facilitates a training program and animal and tissue sharing programs for Center and non-Center members.

    TRAINING AND TISSUE SHARING PROGRAM


  • The Animal Core facilitates the training of any lab personnel from other institutions on techniques required for setting up a rat and mouse intragastric infusion model.

    It also facilitates a tissue sharing program that allows the sharing of tissue specimens obtained from animal models produced by the Core and used by Center investigators.

    NON-PARENCHYMAL LIVER CELL CORE SERVICES

    1. The core will provide freshly isolated or cultured Kupffer cells (KC: hepatic macrophages), hepatic stellate cells (HSC) and sinusoidal endothelial cells (SEC) from normal rats or rat models of liver injury.
    2. The priority of the services will be given to the members of the Southern California Research Center for Alcoholic Liver and Pancreatic Diseases (ALPD) and Cirrhosis.
    3. The services are rendered to the qualified members on a charge back basis to recover the costs of supplies which are not supported by the center and resource grants from NIAAA.
    4. The services can be rendered to non-members for higher charge backs which include personnel costs and surcharges.

    Request for Services

    1. A request for normal rat KC and HSC should be directed to Dr. Kinji Asahina (asahina@usc.edu) and for normal SEC to Dr. Laurie DeLeve (deleve@usc.edu) at least 3 weeks prior to the time the cells are needed.
    2. A request for cell isolation services for the disease models should be made to Dr. Asahina (KC and HSC) or Dr. DeLeve (SEC) at least 2 months in advance.

    a. These charges are based on a whole isolation procedure per rat and not on cell numbers because the cell yield varies. If KC or HSC isolated from one rat are shared, the charges will proportionally be reduced by a fraction. For example, if KC separated from one rat is equally distributed to 2 groups, the charge is $75 for each group of Center members and $100 for each group of Non-members.
    b. In average, 1.8 x 107 KC are isolated from one rat following gradient ultracentrifugation and adherence on plastic dish.
    c. An average yield of freshly isolated HSCs is 3.0 x 107 cells from one rat. After plating on culture dish, an average number of attached HSC is 5.2 x 106 cells per rat.

    MORPHOLOGY CORE SERVICES


    This core provides routine and specialized morphologic services including immunohistochemistry, standardized, blinded histologic assessment, morphometric and image analysis.

    This core provides the centralized administrative services to all city-wide Center components. It also plans and implements public relations and educational programs.
